Effect of Three Anticoagulants on the Cell Characteristics of Autologous Platelet Concentrate in Dogs

Introduction

Different protocols have been described for autologous platelet concentrate (APC) used in regenerative medicine in dogs. And the methodology to concentrate platelets in suprabasal levels differs in the type of anticoagulant used.

Objectives

Describe the effect of three types of anticoagulants (ethylenediaminetetraacetic acid, K3 - EDTA, sodium citrate - SC, citric acid - sodium citrate - dextrose, solution B - ACD-B) on cell characteristics of APCs in canines, obtained using the tube method.

Methods

Venous blood was drawn from twenty-five Brazilian Fila breed dogs, clinically healthy. A sample of blood with each anticoagulant. APC was obtained by the tube method. By volumetric impedance were determined cell characteristics in whole blood and APCs.

Results

The values and statistical inference are summarized in Table 1.

WBC (white blood cell), PCV (packet cell volume), PLT (platelets), LY (lymphocytes), GR (granulocytes), MPV (medium platelet volume), PDW (platelet distribution width). Means with different letters have significant differences by the repeated measures ANOVA.

Conclusions

Anticoagulants used in APCs preparation of canines have influence on cell values of APC. Thus, EDTA has not concentrated platelets and has platelet indices of greater value. The platelet count in the APC is higher with ACD-B than with SC, and no differences between them in the platelet indices are presented.
