Fibrin Tissue Adhesive in the Adherence of Corneal Incision in Rabbits: Scanning Electron Microscopy Study

The purpose of the study was to assess the efficacy of fibrin adhesive for sealing corneal incisions in rabbit eyes. All studies were approved by the institutional Animal Care and Use Committee and were in accordance with the ARVO Statement for the Use of Animals in Ophthalmic and Vision Research. All patients included in this study received a complete ophthalmic examination. The rabbits were anesthetized. All the surgeries were done by a single surgeon. A 3.2 mm clear corneal incision was created with metal knives. Afterwards, the incisions were sealed with fibrin adhesive in group 1 and with a single 10-0 nylon suture in group 2. All eyes were treated with antibiotics and steroids once daily for 15 days. The rabbits were euthanized and the corneas obtained for scanning electron microscopy. Analysis examination 7 and 10 days after surgery. After death the eyes were enucleated and the corneas and a 1.0 mm rim of sclera were excised. Corneas were prepared immediately postmortem for scanning electron microscopy. Photomicrographs at x40 magnification were obtained from each cornea around the incision site using a scanning electron microscope. Using an image analysis software the areas devoid of endothelial cells around the incision site were observed. The results of this study show that corneal endothelial cell loss resulting of the surgical trauma occurred around the incision site in all groups. This study demonstrated that the fibrin tissue adhesives are efficacious in sealing corneal wounds.
