ELISA for Allergen-Specific IgE Determination in Dog. Correlation with in vivo Diagnostics

OBJECTIVES

To study the correlation of the levels of specific IgE against grass pollen allergens, as determined by ELISA, with the results obtained in vivo by intradermal skin tests (ISDT) in a group of grass-allergic dogs.

MATERIALS

Forty-one dogs with clinical histories suggestive of atopy and positive IDST to grasses were selected and classified in three groups according to the intensity of the cutaneous reactions, the number of positive reactions to a standard battery of inhalant allergens (Stallergenes S.A / CBF LETI S.A. ) and the clinical history:

 Group A (17 dogs).- Patients with an atopic pattern of pruritus, with intense seasonal peaks. By IDST, 3 to 4+ reactions to grasses.

 Group B (16 dogs).- Moderate to weak pruritus with seasonal peaks, and 2 to 3+ intradermal reactions to grasses.

 Group C (8 dogs).- Uncertain history of atopic disease. Weak pruritus with no seasonal peaks. By IDST, weak (1 to 2+) reaction to grasses.

 A control group (16 dogs) with negative history of atopy and skin test negative to inhalant allergens was also included. Samples of sera were analyzed for specific IgE against grass pollens using an ELISA method , PET-ELISA® from Alergovet S.L. The results of these analyses were expressed as classes: negative (0), borderline (1), weak positive (2), positive (3) and strong positive (4).

RESULTS

The number of patients with IgE levels within the different classes are indicated for each group of patients.

 Grupo A: 0 (Class 0), 2 (Class 1), 4 (Class 2), 8 (Class 3), 3 (Class 4).

 Grupo B: 3 (Class 0), 3 (Class 1), 7 (Class 2), 1 (Class 3), 2 (Class 4).

 Grupo C: 4 (Class 0), 3 (Class 1), 1 (Class 2), 0 (Class 3), 0 (Class 4).

 Grupo Control: 13 (Class 0), 1 (Class 1), 2 (Class 2), 0 (Class 3), 0 (Class 4).

CONCLUSION

In this study, we have demonstrated that the SAT-ELISA® in vitro test for allergen-specific IgE. in dog has a high degree of sensitivity (83%) and specificity (81%) as compared with the IDST and, in addition, it shows a very good correspondence with the clinical histories of atopy of patients.
