PHAR001-1001: Feeling Inhibited? Experimental Anti-Inflammatory Therapy

COURSE DESCRIPTION
As a foundation for understanding the role therapeutic agents, the following aspect of the inflammatory processes will be discussed:

• Cellular death with production of inflammatory mediators
• Cellular death (necrosis) vs programmed cell death (apoptosis)
• Activation of apoptosis via capase dependent pathways
• Prostaglandin synthesis via the cyclooxygenase pathways
• TNF family of cytokines
• Roles of MMP (matrix metalloproteinases) in disease

With that background the following agents and their therapeutic potential will be discussed:

• Treatment to reduce effects of prostaglandin damage
• Non-selective (Cox 1 & Cox 2) vs Cox 2 selective NSAIDS therapies
• Experimental therapies using NSAIDS in cancer (anti-angiogenesis effects)
• Leukotriene synthesis and their role in inflammation
• Experimental therapy using leukotriene blockers in inflammatory disease such as asthma, rhinitis, polyps, interstitial cystitis and bowel disease
• TNF alpha role in disease such as heart failure, shock, cancer
• The specific family of TNF called TRAIL (tumor-necrosis-faction-related-apoptosis-inducing-ligand)
• TNF alpha antagonists and their experimental use in cancer in relation to their anti-angiogeneis effect
• Relationship between MMP and TIMP (tissue inhibitors of matrix metalloproteinases) Specific
• Use of MMP inhibitors in diseases like peridontal disease, arthritis and cancer will be reviewed

At the end of this course, participants will have a better understanding of the pathophysiology of inflammation the potential roles of several new therapeutic options being developed including TNF alpha antagonists, MMP inhibitors, Prostaglandin antagonists (NSAIDS) and Leukotriene blockers.
